Universal T Cell Assay Standard TERS™ Kit
Generate MHC-class-I restricted T-cell receptor (TCR)-engineered reference samples containing defined numbers of functional, antigen-specific T cells for standardization and monitoring of:

  • MHC-Multimer staining (e.g. Dextramer ®);
  • intracellular cytokine detection (ICS);
  • ELISPOT assays.

They represent an ideal tool for validating and monitoring assay performance within or across institutions. This new product may be used to standardize antigen-specific tests over time, between different platforms, or different laboratories. It may also be used to calibrate antigen-specific assays.

Specificity
TERS™ Kits can be used to monitor the performance of functional and non-functional T-cell assays irrespective of the specificity selected. They are run in the same way as usual assay standards, i.e. in parallel to other samples tested in a given assay run. Please note, that for functional testing involving T-cell stimulation by a peptide presented on a specific MHC molecule, the MHC of the used cell materials must match the MHC restriction of the TERS Kit as indicated. In the event that the peptide recognized by the TERS is the same as used for stimulating other samples in an assay run, an additional level of control (e.g. integrity of the specific peptide) is provided. If this is desired, you may wish to check if a Kit for producing TERS recognizing your specific peptide is available

The preparation of a TERS™ batch with this product requires peripheral blood mononuclear cells (PBMCs), which may be obtained from a human buffy coat, for example (not supplied). Additional reagents, an electroporator and a flow-cytometer are required during the production process.
